Περίληψη: Background: Neutrophil extracellular traps (NETs) are implicated in a variety of pathophysiological conditions, including atherogenesis and thrombosis. Platelet-derived microparticles (PMPs) are membrane-shedded vesicles that may also play important roles in the pathogenesis of various disorders. Aims: We investigated the effect of activated platelets and PMPs on NETs formation (NETosis), in vitro. Methods: Neutrophils (200,000) were isolated from whole blood of healthy volunteers, attached to poly-L-lysine coverslips and incubated with autologous platelet-rich plasma (PRP), either in its resting state, or pre-activated in an aggregometer with 20μM ADP, or activated in situ in the presence of neutrophils with 50μM ADP. In other experiments, PMPs were prepared from washed platelets activated with 0.2U/mL thrombin. PMPs were isolated by ultracentrifugation, and incubated with neutrophils at concentrations ranging from 1-60μg protein/mL. In all experiments, neutrophils were incubated for 3.5h at 37℃, 5% CO2, stained, and the %NETosis was microscopically determined. Results: ADP-activated PRP increased NETosis, by 106±32%, compared to untreated samples (pre-activation in the aggregometer), p=0.03 and by 63±22% (in situ activation), p=0.011. PMPs increased NETosis in a dose-dependent manner, exhibiting an EC50 value of 9.4μg protein/mL. Conclusions: ADP-activated platelets are potent inducers of NETosis, an effect that could be at least partially attributed to PMPs. Thus, inhibiting platelet activation, and targeting PMPs, could offer novel therapeutic approaches in preventing various pathophysiological conditions in which NETs are implicated, including atherosclerosis and thrombosis.
